Changes in immunocompetent cells in damaged muscle tissue following eccentric muscle contraction exercise
ABSTRACT
The purpose of this study was to examine immunocompetent cells in damaged muscle tissue following eccentric muscle contraction. Eight-week-old Institute of Cancer Research (ICR) mice (n=126) were divided into 3 groups: high frequency group (HG, n=42), middle frequency group (MG, n=42), and low frequency group (LG, n=42). The tibialis anterior muscle was stimulated via the peroneal nerve to contract eccentrically muscle exercise (HG; 90 times × 4 sets, MG; 30 times × 5sets, LG; 10times × 5sets, Rest for 1 minute between each set). The tibialis anterior muscle was isolated before (n=6) and 1 (n=6), 3 (n=6), 7 (n=6), 14 (n=6), and 28 d (n=6) after muscle contraction. Neutrophils were significantly increased in all groups after 3 days of muscle contraction (p<.05) compared with before of muscle contraction. Neutrophils in LG and HG were significantly increased after 1 day and 7 days of muscle contraction (p<.05) compared with before of muscle contraction. Macrophages and cytotoxic T cells were significantly increased in all groups immediately after, 1, 3, 7, 14, and 28 days of muscle contraction (p<.05) compared with before of muscle contraction. After 3 days of muscle contraction, cytotoxic T cells were significantly higher (p<.05) in the MG and HG compared with LG. In addition, IL-6 analysis showed an expression in 14 days (LG), 3days (MG), and 7 days (HG) after muscle contraction (P <.05). These results demonstrated that eccentric muscle contraction exercise increase immunocompetent cells in damaged muscle tissue, and IL-6 gene expression.
