ABSTRACT |
The purpose of this study was to examine immunocompetent cells in damaged muscle tissue following eccentric muscle contraction. Eight-week-old Institute of Cancer Research (ICR) mice (n=126) were divided into 3 groups: high frequency group (HG, n=42), middle frequency group (MG, n=42), and low frequency group (LG, n=42). The tibialis anterior muscle was stimulated via the peroneal nerve to contract eccentrically muscle exercise (HG; 90 times × 4 sets, MG; 30 times × 5sets, LG; 10times × 5sets, Rest for 1 minute between each set). The tibialis anterior muscle was isolated before (n=6) and 1 (n=6), 3 (n=6), 7 (n=6), 14 (n=6), and 28 d (n=6) after muscle contraction. Neutrophils were significantly increased in all groups after 3 days of muscle contraction (p<.05) compared with before of muscle contraction. Neutrophils in LG and HG were significantly increased after 1 day and 7 days of muscle contraction (p<.05) compared with before of muscle contraction. Macrophages and cytotoxic T cells were significantly increased in all groups immediately after, 1, 3, 7, 14, and 28 days of muscle contraction (p<.05) compared with before of muscle contraction. After 3 days of muscle contraction, cytotoxic T cells were significantly higher (p<.05) in the MG and HG compared with LG. In addition, IL-6 analysis showed an expression in 14 days (LG), 3days (MG), and 7 days (HG) after muscle contraction (P <.05). These results demonstrated that eccentric muscle contraction exercise increase immunocompetent cells in damaged muscle tissue, and IL-6 gene expression. |
