혈관기능 항상성을 위한 유산소성 운동과 미토콘드리아

Abstract

PURPOSE

The purpose of this review was to determine the role of mitochondria in the maintenance of vascular homeostasis and to investigate the benefit of exercise in the regulation of mitochondrial integrity.

METHODS

In this review, a literature review was performed to summarize the role of mitochondria in the functional maintenance of vasculature and the effect of aerobic exercise training on the mitochondrial quantity- and quality-control.

RESULTS

Enhancing structural and functional integrity of mitochondria is regarded as a novel therapeutic target for the prevention and the improvement of cardiovascular diseases. Mitochondria have been regarded as the intracellular power plant for the generation of ATP, the molecular resource of intracellular energy transfer. However, the role of mitochondria goes beyond the oxidative phosphorylation in the vasculature. Mitochondrial dysfunction is directly associated with the development of cardiovascular diseases, such as atherosclerosis, hypertension, hyperlipidemia, and diabetes, by enhancing pro-inflammatory, pro-apoptotic, and pro-senescent mechanisms. Regular training of aerobic exercise may contribute to the maintenance of vascular homeostasis by regulating mitochondrial biogenesis, dynamics, and clearance.

CONCLUSIONS

Mitochondria are the emerging therapeutic target of the aerobic exercise for the maintenance and the improvement of vascular homeostasis.

서 론

심혈관 질환은 서구사회에서 전체 사망률의 가장 높은 비율을 차지하고 있으며, 그 중 2/3에 해당하는 원인은 죽상동맥경화(atherosclerosis)와 직접적인 관련이 있는 것으로 나타나고 있다[1]. 이러한 심혈관 질환 및 동맥경화는 이제 서구사회뿐만 아니라, 서구식 식단을 즐겨하는 한국 사회에서도 주요 사망원인으로 그 위험성이 증가하는 것으로 보고되고 있다[2]. 동맥경화 플라크(atherosclerotic plaque)는 혈관 평활근 세포(vascular smooth muscle cells), 단핵 백혈구(monocyte), 대식세포(macrophage), T림프구 등과 같은 면역 세포들과 세포 내외의 지질 및 세포 파편들을 포함하는 다양한 세포들의 병변을 일컫는다. 저밀도 지단백(low-density lipoprotein, LDL)과 같은 혈중 지질 성분의 증가는 동맥경화 플라크의 축적을 증가시키는 주요 위험 요인이 된다[3]. 혈중 LDL이 혈관내 염증 부위 또는 혈류가 원활하게 생성되지 않는 특정 부위로 이동하여, 지질산화효소(lipoxygenase) 및 활성산소종(reactive oxygen species, ROS)과 반응하고 이로 인한 LDL 산화작용과 혈관 내피세포(endothelial cells)의 기능이상은 동맥경화를 유발하는 주요 기전으로 작용한다[4,5]. 플라크는 일반적으로 혈관 내피세포의 기능이상으로 인해 혈관 내 혈액의 흐름과 전단응력(shear stress)이 정상적으로 생성되지 않는 부위에서 축적되고, 백혈구(leukocyte), 혈소판(platelet), 접합 분자(adhesion molecules)와의 결합성 증가와 함께 성장하며 그 염증을 악화시킨다[6]. 이는 혈관 내 ROS 생성 및 DNA 손상을 증가시키며, 세포 노쇠(senescence)와 세포 사멸(apoptosis)을 증진시킨다. 이러한 혈관 세포 내 염증 반응, 노쇠, 세포사(cell death)가 혈관 내 병변을 유발하고, 악화 시 파열에 이르게 한다[7].
미토콘드리아는 흔히 ‘세포 내 에너지 발전소’라 지칭되며, 세포 내 에너지 수송 분자인 ATP 생산의 주요 세포소기관으로 기능한다[8]. 그러나 미토콘드리아의 기능은 에너지 생산뿐 아니라 ROS 생성과 조절, 칼슘의 생성과 조절, 세포 사멸의 조절 등에 이르기까지 그 기능이 다양한 것으로 알려져 있다[9]. 또한 최근, 유산소성 운동에 따른 혈관 내 전단응력의 증가와 이로 인산 미토콘드리아 신생(mitochondrial biogenesis)이 혈관의 염증 수준과 노쇠를 억제·개선하였다는 연구들이 보고되고 있으며[10,11], 미토콘드리아의 유산소성 운동을 통한 혈관 기능 항상성 유지와 혈관 질환 억제에 핵심적인 기전으로서의 가능성을 시사하였다[12]. 따라서 본 연구에서는 유산소성 운동에 따른 혈관세포 내 미토콘드리아의 양적·질적 향상이 혈관 기능의 항상성 유지와 개선에 어떠한 잠재적 역할을 담당하는지에 대해 그동안의 선행연구들을 통해 고찰하고, 유산소성 운동과 혈관 건강에 있어서 미토콘드리아의 역할과 의미에 대해 종합·정리하고자 한다.

본 론

1. 혈관에서의 미토콘드리아 기능 및 특성

1) 혈관 미토콘드리아 볼륨

혈관세포 미토콘드리아의 볼륨은 많은 에너지를 필요로 하는 다른 세포와 비교해서 상대적으로 적은 편에 속한다. 동물 실험에서 밝혀진 연구결과에 따르면, 혈관 미토콘드리아는 세포 전체 볼륨의 약 6%를 차지하는 것으로 알려져 있다. 이에 반해 간세포에서는 약 28%, 심장근 세포에서는 32%의 볼륨을 차지하는 것으로 알려져 있다[13,14]. 상대적으로 낮은 수준의 볼륨은 미토콘드리아가 혈관에서 산화적 인산화를 통한 에너지 생성에 많은 기여를 하지 않는 것을 나타낸다[15]. 이에 따라 혈관세포는 상당량의 에너지를 무산소성 해당과정을 통하여 에너지를 획득하게 된다. 관상동맥에서 추출한 In vitro 실험에서 밝혀진 연구에 따르면, 혈관세포에서 약 99%의 포도당은 산화적 인산화를 거치지 않고 무산소성 해당과정의 결과물인 젖산으로 변하며, 미토콘드리아의 산소호흡은 주로 세포에 내재하고 있는 대사산물 산화에 국한되는 것으로 밝혀졌다[16]. 이에 따라서, 혈관미토콘드리아의 주요 역할은 에너지 생성에 기여하는 것보다 혈관의 상태를 감시하고 조절하는 세포 내 필수 신호전달 기관으로 강조되고 있다[15,17].

2) 미토콘드리아의 혈관세포 내 위치

일반적으로 미토콘드리아의 세포 내 위치는 에너지 생성을 위한 효율성에 영향을 미칠 뿐 아니라, 세포 내 다른 소기관들과의 신호전달에도 매우 중요한 역할을 담당한다[15]. 혈관 미토콘드리아는 세포의 형태를 유지시키는 데 필요한 세포골격단백질(cytoskeleton)에 연결되어 있으며, 혈액 흐름으로 유도되는 전단응력에 의해 그 기능이 변화된다. 또한 세포 내외부의 다양한 자극에 반응하여 미토콘드리아 ROS를 발생시키고, 이를 통해 다른 소기관들과 신호를 전달하는 것으로 알려져 있다[18]. 혈관 세포가 산소부족 상태에 놓이게 되면, 미토콘드리아 주변의 운동성단백질인 dynein을 활성화시켜 미토콘드리아를 핵 주변으로 이동시켜 핵과의 신호 교신 효율성을 조절하게 된다[19]. 이와 같이, 혈관세포 내 미토콘드리아는 정적인 기관이 아니며, 세포 내외부의 자극에 따라 활발하게 이동하면서 신호전달자 역할을 한다. 추가적으로, 미토콘드리아는 칼슘이온의 저장소 역할을 하는 소포체와 인접하여 칼슘이온의 중화작용을 향상시킴으로써, 혈관 세포 내 칼슘이온농도의 항상성 유지에도 역할을 담당한다[20]. 실제적으로 소포체와 미토콘드리아의 거리는 미토콘드리아의 막전위와 세포사멸을 조절하는 데 밀접한 연관이 있는 것으로 알려져 있다[21].

3) 혈관 미토콘드리아의 ROS 생성 및 조절

미토콘드리아는 ROS가 생성되는 주요 소기관으로 알려져 있으나, 이를 중화하는 기능도 가지고 있다. 따라서 미토콘드리아는 활성산소종의 미세조절로 혈관세포의 기능을 통제하는 것으로 알려져 있다. 미토콘드리아의 기작으로 에너지가 생성되는 과정에서 발생되는 전자는 전자 전달계(electron transport chain)를 거쳐 다섯 번째 미토콘드리아 효소인 제5복합체(mitochondrial Complex V)에 도착하게 된다[22]. 이 과정에서 전체 환원되는 전자 중 1-3%는 산소분자와 불완전한 결합을 하게 되어 과산화물(superoxide)과 과산화수소(H2O2, hydrogen peroxide) 등을 생성하는데, 이를 총칭하여 미토콘드리아 ROS (mtROS)라 정의한다[23]. 관련 연구에 따르면 대다수의 미토콘드리아 ROS는 complex I과 complex III에서 주로 생성되는 것으로 확인된다[13]. 최근 전자 전달계에서 생성되는 ROS뿐만 아니라 다른 요인에서 발생되는 ROS도 최근 활발히 연구되고 있다. 대표적인 분자로는 Nox4 (NADPH oxidase 4)로 알려져 있는데 미토콘드리아 내에 많은 양이 발현되며, 혈관세포의 증식에도 깊은 연관이 있는 것으로 알려져 있다. 특히 Nox4는 혈관 염증반응 시에 과산화물보다 과산화수소를 주로 발생시키며 혈관세포를 보호한다는 연구결과가 보고되어, 생리학적 수준 내의 ROS는 세포생존의 중요한 신호로 작용할 수 있음을 보여준다 [24]. 생리학적 수준을 초과하는 양의 미토콘드리아 ROS가 발생되면 이를 빠르게 중화시키는 기전이 작동한다. Manganese-dependent superoxide dismutase (MnSOD)는 미토콘드리아에서 가장 많이 발현되는 항산화효소로 알려져 있으며, 이는 미토콘드리아 기질에서 과산화물을 중화시키며, copper zinc superoxide dismutase (CuZnSOD)는 미토콘드리아 막 외부로 이동하는 과산화물을 과산화수소로 환원시킨다[25]. 과산화수소는 과산화물에 비해 반감기가 길고 비교적 안정적이지만 추가적인 환원이 필요하다. 이를 관장하는 항산화효소는 카탈라아제(catalase)와 페록시다아제(peroxidases)가 있다. 카탈라아제는 주로 세포질 퍼옥시좀(peroxisomes)에 존재하며, 미토콘드리아 내에 존재하는 티오레독신-2 (thioredoxin-2), 페록지디옥신-3 (peroxidioxin-3), 글루타레옥신-2 (glutaredoxin-2), 그리고 글루타시온페록시다제-1 (glutathione peroxidase-1) 등이 있다[15]. 해당 항산화 효소들은 과산화수소를 생명체에 무해한 물로 환원시켜 세포 생장 과정을 조절한다.

2. 미토콘드리아 손상과 혈관 질환

1) 이상지질혈증과 동맥경화

동맥경화 유병에 있어서 혈관 세포의 미토콘드리아 손상과 기능장애가 나타난다는 사실은 여러 임상 연구와 동물 실험에서 그 증거들이 나타나고 있다. 동맥경화 질환자의 혈관 평활근 층에서는 미토콘드리아 DNA (mitochondrial DNA)의 결손이 두드러지며, 이러한 현상은 혈관 세포가 ROS에 노출됨에 따라 발생하여 산화적 손상을 일으키고 결과적으로 미토콘드리아 단백질의 발현 저하와 ATP 생산능력 감소를 유발하게 된다[26,27].
고지혈증(hyperlipidemia)은 동맥경화의 위험 인자로 잘 알려져 있으며, 아포지질단백질 E (ApoE)는 지질 입자들이 세포 조직 내로 흡수되기 위한 필수 구성성분으로 기능한다. ApoE 결손 쥐(ApoE-/-mice)에게서는 고지혈증과 이후 동맥경화로의 진행이 두드러지게 나타나는데, 미토콘드리아 DNA의 결손은 이러한 동맥경화로의 진행에 선행하여 필수적으로 나타나는 것으로 보고되고 있다[28]. 이러한 현상은 항산화 효소의 활성이 억제된 상황에서 더욱 악화되는 것으로 나타났다[28]. 흡연은 동맥세포 내에서 이러한 미토콘드리아 DNA의 손상과 ATP 생산 저하를 통한 동맥경화 유발의 가장 대표적 위험 인자로 잘 알려져 있다[29]. 최근의 연구에서는 ApoE 결손 쥐에게서 핵 DNA 및 미토콘드리아 DNA의 손상이 함께 발생하였으며, 미토콘드리아 제1복합체(mitochondrial Complex I)의 손상에 의한 미토콘드리아 호흡사슬복합체(mitochondrial respiratory chain) 질환과, 이를 통한 동맥경화의 유발이 가속되는 것으로 보고되고 있다[30]. 미토콘드리아 호흡사슬복합체에서 최종적인 ATP 생산에 직접적으로 기능하는 것은 ATP 합성효소(ATP synthase)인 미토콘드리아 제5복합체(mitochondrial Complex V)이지만, 제1, 3, 4 복합체 또한 미토콘드리아 전자전달계(electron transport chain)에 의한 막 전위차 형성과 미토콘드리아 호흡에 직접적으로 기능하는 요소들로써, 미토콘드리아 호흡사슬복합체 일부의 손상은 결과적으로 미토콘드리아 전체의 기능 손상과 혈관 세포 기능 저하를 유발하게 되는 것이다[31,32]. 아울러, 혈관에서의 미토콘드리아 DNA 손상은 죽종 형성(atherogenesis)을 통한 각종 심혈관 질환의 유발과도 밀접하게 관련되어 있는 것으로 보고되고 있다[33].

2) 혈관의 염증과 노화

혈관 내피세포의 활성화(endothelial activation)란 안정기에 있던 내피세포층이 외부에서 주어진 스트레스 또는 혈관세포 내부로 침습된 병변에 의해 백혈구와의 반응성이 증가하고, 혈소판 및 접합 분자들(adhesion molecules)의 결합 숫자가 증가하는 등 혈관 내부의 방어 기전이 증가하는 염증전(pro-inflammatory) 단계의 상태를 의미한다[34]. 고혈압, 당뇨, 비만, 신체적 비활동, 고 콜레스테롤, 흡연 등과 같은 심혈관 질환 위험 인자들은 이러한 내피세포의 활성, 즉 혈관 염증반응의 유발을 증가시키는 것으로 알려져 있으며, 이는 혈관 노화와 관련이 깊은 질환인 동맥경화의 초기단계적 특성으로 인식되고 있다[35,36]. 안정기의 혈관 내피세포는 산화질소(nitric oxide, NO)의 생성에 의해 혈관의 수축 ·이완 기능이 유지되고, 혈전(thrombosis)의 침착이 억제되며, 내피세포의 증식이 조절되는 등 혈관기능의 항상성이 유지된다[37]. 또한 NO는 혈관 내피세포 내 미토콘드리아의 산화적 인산화를 조절하고 ROS의 생성을 억제하는 등 혈관 내피세포의 안정상태를 유지하는 데 기여한다[38]. 그러나 과산화수소와 같은 ROS의 생성과 세포 전체를 통한 확산은 미토콘드리아와 반응하여 세포 기능을 변화시키며, NO 생성에 촉매작용을 통해 혈관 기능 항상성에 핵심적인 역할을 담당하는 혈관 내피세포 산화질소 합성효소(endothelial nitric oxide synthase, eNOS)가 오히려 과산화물 생성을 유발하게 하는 등, 염증반응 활성상태로 혈관 내피세포 내부의 환경을 변화시키게 된다[39,40]. 이렇듯 ROS와 NO의 상호작용은 혈관 내 염증반응을 불러일으키고, 반복된 염증반응의 악순환은 동맥경화와 같은 혈관 노화성 질환을 유발하게 되는 것이다[41]. 고혈압, 비만, 운동 부족, 흡연 등과 같은 심혈관 질환 위험 요인에 대한 지속적인 노출과 세포내 항산화효소(antioxidant enzyme)의 기능을 초과하는 만성적인 ROS의 과도한 생성은 혈관 내피세포의 항염증성 기능을 저하하고, 내피세포층의 박리를 유발하게 된다[42]. 이러한 내피세포층의 결속력 저하와 박리현상은 혈관 내 염증 반응을 악화시키고, 이 상태가 지속될 경우, 이른 시기에 혈관 노화가 발생하고 동맥경화와 같은 노인성 혈관 질환에 노출되게 되는 것이다. 염증반응이 활성화된 혈관 내피세포 또는 세포사멸 반응이 진행 중인 내피세포에서 생성·방출되어 혈액 중에 순환하는 endothelial microparticles (EMP)의 수치는 이러한 혈관 내피세포의 염증 상태와 혈관 노화 상태를 알려주는 생체지표(biomarker)로 알려져 있다[43,44]. 혈중 EMP는 동맥경화, 고혈압, 관상동맥질환자, 당뇨와 같은 심혈관계 질환자에게서 그 생성량이 매우 높은데[45], 규칙적인 유산소성 운동 훈련을 통한 혈관 내피세포의 미토콘드리아의 증가는 이러한 혈관 염증반응의 지표인 EMP의 생성량을 감소하는 것으로 보고되고 있다[10]. 즉, 체내 ROS의 증가와 미토콘드리아 기능 이상에 의해 유발되는 혈관 염증반응과 이로 인한 혈관노화는 동맥경화와 같은 혈관 질환을 유발할 수 있으며, 이는 규칙적인 운동을 통한 미토콘드리아 증가에 의해 억제될 수 있는 것으로 밝혀졌다.

3) 고혈압과 당뇨

고혈압은 죽상동맥경화 유발의 위험요인이며, 미토콘드리아의 에너지 결핍과 칼슘 이온 과부하와 같은 기능 이상은 동맥의 혈압 증가와 고혈압의 만성적 발병에 직접적으로 관련이 된다[46]. 고혈압 유발 쥐를 이용한 실험에서 나타난 바에 의하면, 미토콘드리아의 에너지 손상과 칼슘 과부하는 쥐 심장근의 이상적인 비대를 유발하는데, 이러한 심장근비대가 고혈압 발생의 주요 원인이 되는 것으로 나타났다[47,48]. 미토콘드리아의 항산화 시스템도 고혈압으로부터의 보호에 있어 매우 중요한 기전이 된다. 미토콘드리아에서 생성되는 주요 항산화효소인 MnSOD의 결핍 쥐에서는 노화와 고염분 식이에 의해 동맥 혈압이 크게 증가하였으며, 동맥경화가 유발되는 것으로 보고되고 있다[49]. 즉, 미토콘드리아의 항산화 시스템 이상 자체가 고혈압 유발의 원인이 될 수 있음이 드러난 것이다. 최근의 연구에서는 미토콘드리아 tRNA의 변이가 고혈압, 고콜레스테롤 혈증, 혈중 마그네슘 결핍증을 유발한 것으로 보고되고 있다[50]. 이러한 미토콘드리아 기능 이상 환자들에게서는 약 30대에 혈중 콜레스테롤 농도와 혈압의 증가가 나타나는 것으로 관찰되었는데, 이는 미토콘드리아의 유전적 이상과 이로 인한 기능 저하 자체가 고혈압과 고콜레스테롤 혈증을 유발하는 요인으로 작용한다는 것을 보여준다고 하겠다[51].
당뇨는 관상 동맥 질환과 관련된 사망률 증가의 주요 위험 요인이며[52,53], 특히 2형 당뇨 환자들에게서는 심근 허혈 빈도가 더욱 높게 나타나며, 심근경색 증세와 이로 인한 사망률이 매우 높은 것으로 알려진다[54]. 전 세계적으로 이러한 당뇨 발병률은 계속해서 증가하는 추세에 있으며, 따라서 당뇨 발병의 기전에 대한 이해를 위한 노력은 지속되고 있다. 인슐린 저항성은 2형 당뇨 및 당내성 손상을 가진 환자에게서 나타나는 주요 현상이다[55]. 인슐린 저항성은 혈중 포도당을 근육 및 지방 조직으로 수송하는 능력의 저하로 인해 만성적인 고혈당 증세를 유발하게 된다[56,57]. 이러한 지속적인 고혈당 증세는 혈관 내피세포 미토콘드리아 전자전달계에서의 과산화물(superoxide) 생성을 촉진하게 되며, 이로 인한 산화 스트레스는 결국 혈관 기능의 손상을 일으키게 된다[58]. UCP1 또는 MnSOD의 과발현 등을 통해 미토콘드리아 생성 ROS의 수준을 낮추었을 때, 동맥경화와 같은 고혈당 유발성 혈관 기능 손상의 기전적 원인이 되는 protein kinase C (PKC)의 활성 및 advanced glycation end products (AGE)의 생성 억제, nuclear factor kappa B (NfkB)의 과축적을 유발하는 폴리올 대사계의 활성 억제가 나타났다[59]. 혈관 내피세포에서의 NfkB의 활성은 vascular cell adhesion molecule-1 (VCAM-1) 및 monocyte chemoattractant protein-1 (MCP-1)과 같은 세포 접합 분자의 발현을 유도하며, 이는 결국 혈관 내 염증반응 및 동맥경화를 유발하는 것으로 알려져 있다[60]. 즉, 당뇨로 인한 지속적인 고혈당 상태는 혈관 내피세포 미토콘드리아의 과부하로 인한 기능 손상과 이로 인한 미토콘드리아 ROS 생성 증가를 유발하고, 이는 결국 혈관 기능의 장애를 발생시키는 원인이 되는 것이다. 규칙적인 유산소성 운동이 인슐린 민감도 증가를 통해 당뇨의 개선을 유발한다는 사실을 고려하였을 때, 유산소성 운동을 통한 당처리 능력의 향상이 결국 혈관 내피세포의 미토콘드리아 기능 향상은 물론, 혈관 기능 개선을 위한 궁극적인 해결책이 될 수 있겠다[61].

3. 유산소성 운동과 혈관 미토콘드리아

1) 운동과 미토콘드리아 신생(mitochondrial biogenesis)

생체 내 미토콘드리아는 생성과 제거를 끊임없이 반복하며 균형 잡힌 상태로 유지된다. 이 과정에서 미토콘드리아 DNA가 복제되고 핵과 미토콘드리아 내에 있는 유전자들이 발현된다. 이 중 가장 잘 알려진 분자는 미토콘드리아 신생의 통제자라고 불리는 peroxisome proliferation-activated receptor γ co-activator 1α (PGC-1α)가 있다. PGC-1α는 전사조절 보조활성제로서 미토콘드리아의 신생과 에너지 대사 조절에 중추적인 역할을 담당한다[62]. 유산소성 트레이닝은 PGC-1α의 양을 증가시키는 것으로 잘 알려져 있는데, 최근 연구에서는 일회성 운동만으로도 전사 및 번역과정이 증가되는 것을 보고하여 운동의 중요성이 강조되고 있다[15,63]. 이러한 결과는 PGC-1α를 전사하기 위한 상위 신호전달 구성체는 운동으로 유도되는 스트레스 및 전단응력을 신속히 감지하는 것을 의미한다. 이와 관련하여, 운동성 미토콘드리아 신생은 관련 유전자 전사에 앞서 PGC-1 α의 핵 내 이동으로 시작된다[64]. 추가적으로 일회성운동은 핵내뿐만 아니라 미토콘드리아 내의 PGC-1α의 양도 증가시킨다[65]. 이렇게 증가된 PGC-1α는 핵 내에서는 NRF-1, 2와 결합하고, 미토콘드리아 내에서는 TFAM (mitochondrial transcription factor A)과 결합하여 핵과 미토콘드리아 내의 유전자들을 개별적으로 전사시킬 뿐 아니라, 미토콘드리아 DNA의 복제도 유도한다. PGC-1α는 인산화(phosphorylation)와 탈아세틸화(deacetylation)를 거치면서 활성도가 높아진다[62].

2) 운동과 미토콘드리아 다이나믹스(mitochondrial dynamics)

미토콘드리아는 활발히 움직이는 세포 소기관으로 알려져 있는데 신호에 따라 서로 융합(fusion)하기도 하고 분열(fission)도 한다. 이러한 융합과 분열을 통칭하여 미토콘드리아 다이나믹스라 한다. 이 과정에는 많은 신호단백질이 관여하는데, mitofusins 1 and 2 (MFN1 and MFN2)는 미토콘드리아의 외막의 융합에 관여를 하며 optic atrophy type 1 (Opa1)은 미토콘드리아 내막의 융합을 유도하는 것으로 알려져 있다[66]. 미토콘드리아 외막의 분열은 dynamin-related protein 1 (Drp1)와 Fission 1 (Fis1)에 의해 조절되는 것으로 알려져 있다[67]. 최근 연구에서 미토콘드리아의 다이나믹스가 혈관세포의 미토콘드리아기능뿐만 아니라 세포사멸 및 노화에도 깊은 영향에도 있다는 결과가 보고되고 있다[68]. 이와 연관하여 미토콘드리아 다이나믹스의 결핍은 혈관세포의 기능을 저하시켜 죽상 경화증 및 고혈압에 부정적인 영향을 미친다[13,69].
운동은 미토콘드리아 분열과 융합 모두를 조절하는 것으로 알려져 있다. 특히, 장기간의 유산소성 운동은 MFN1과 Fis1의 단백질 양을 증가시키는 것으로 나타났으며[70], 일회성 운동 후 24시간이 지난 뒤 MFN1과 MFN2의 mRNA 전사가 증가됨이 보고되었다[71]. 흥미롭게도, PGC-1α와 estrogen-related receptor α (ERRα)는 운동 후 2시간 뒤 빠르게 mRNA 전사가 이루어졌다[71]. MFN1과 MFN2의 전사는 PGC-1α과 ERRα의 상호작용으로 유도되는 것을 확인해 볼 때[72], 운동으로 유도되는 PGC-1α는 미토콘드리아의 신생뿐만 아니라, 다이나믹스에도 영향을 미칠 것으로 사료된다.

결 론

혈관 세포의 미토콘드리아의 양은 체내 다른 세포보다 상대적으로 적은 양으로 이루어져 있으며, 대다수의 에너지를 무산소성 해당과정으로 충당한다. 이러한 사실은 혈관 세포 내의 미토콘드리아는 에너지를 생산하는 데 일차적인 목표가 있는 것이 아니라 세포의 항상성과 기능을 조절하는 통제자 역할을 하는 것으로 사료된다. 손상받은 미토콘드리아는 생리학적인 수준을 초과하는 ROS를 발생시켜 혈관세포 노화를 유도하여 죽상 경화증 및 고혈합 등을 발생시킨다. 따라서 미토콘드리아의 질은 혈관 건강에 직결된다. 유산소성 운동은 혈관세포의 건강유지에 아주 효과적이며[73-75], 미토콘드리아의 신생(biogenesis) 및 다이나믹스(dynamics)를 조절하는 것으로 알려져 있다. 균형 잡힌 신생, 융합, 분열을 통한 미토콘드리아 재건과정은 혈관세포의 손상된 미토콘드리아를 제거하고 대체한다. 따라서 운동은 미토콘드리아의 양뿐만 아니라 질까지도 조절하며 혈관의 항상성 유지에 지대한 영향을 미친다.
