The Effect of Endurance Exercise Training on Inflammatory Cytokines, Apoptosis, and FOXO Expression in Adipose Tissue of Hypertensive Rats |

ABSTRACT |
The purpose of this study was to investigate the expression of p53, FOXO1, FOXO3a, inflammatory cytokines and apoptosis in adipose tissue of endurance exercise trained rats. The Wistar-Kyoto rats (WKY) and spontaneously hypertensive rats (SHR) were divided into 3 groups: WKY control (WKY, n=9), SHR control (SHR-C, n=9), SHR exercise (SHR-E, n=9), and submitted to treadmill exercise for 16 weeks (16 m/min, 5 days/week, 30-60 min/day). Every other weeks, systolic blood pressure were measured. After the last bout of exercise, all rats were sacrificed, then, p53, FOXO1, and FOXO3a were analyzed by western blotting. Also, inflammatory cytokines (IL-6, TNF-α, MCP-1) and apoptotic proteins(Bax, Caspase-9, and Caspase-3) were analyzed by RT-PCR. The expression of inflammatory cytokines (TNF-α, MCP-1) and apoptosis markers (Bax, Caspase-9, and Caspase-3) were significantly decreased in SHR-E compared to the SHR-C. Additionally, p53 was significantly decreased and FOXO1 was significantly increased by exercise in SHR rats. These findings suggest that endurance exercise may decreases systolic blood pressure via enhancement of FOXO1 and suppression of inflammatory cytokines (TNF-α, MCP-1) and apoptosis factors (p53, Bax, Caspase-9, and Caspase-3) in adipose tissue of SHRs. |
